Buffalo Bill and Wild Bill Hickok join forces to establish a mail route that can get mail from St. Joseph, Missouri, to Sacramento, California, in ten days. Along the way they must battle bad weather, hostile Indians and outlaws intent on robbing the mail and shutting down the entire operation.

The film starts off with an old man in the desert and two signs are shown; one that says East, the other says west. Japan has sent a mission to San Francisco. In San Francisco, the Japanese are surprised by the American culture. One American, Gus Taylor, and his gang steal all the gold from the mission and make their way in to the desert. One of the samurai of the mission chases after the gang into the desert. He is joined by a young American boy, Sam, whose father was killed by the gang leader. The group picks up a variety of people along the way to New Mexico. The Japanese and the Americans on the trip share parts of their own cultures with each other. The group of vigilantes eventually makes it to New Mexico and finds the gang. They take back their stolen gold and return to San Francisco.

When the Pony Express disbands, riders Tennessee and Johnny head for Adobe Wells. Tennessee becomes the Deputy Marshal while Johnny joins an outlaw gang. But to no avail as Johnny murders a man and this time Tennessee must do his duty.

The story centres around Jonathon Tibbs, the young son of a family of English gunsmiths. He travels to the American West in the 1880s to sell firearms to the locals. While there he inadvertently acquires a reputation of quickness on the draw due to his wrist mounted Derringer style weapon and from there the post of sheriff. He endeavours to clean up the town using what skills he has and by multilateral diplomacy.
Embezzler, shill, all around confidence man S. Quentin Quale is heading west to find his fortune; he meets the crafty but simple brothers Joseph and Rusty Panello in a train station, where they steal all his money. They're heading west, too, because they've heard you can just pick the gold off the ground. Once there, they befriend an old miner named Dan Wilson whose property, Dead Man's Gulch, has no gold. They loan him their last ten dollars so he can go start life anew, and for collateral, he gives them the deed to the Gulch. Unbeknownst to Wilson, the son of his longtime rival, Terry Turner (who's also in love with his daughter, Eva), has contacted the railroad to arrange for them to build through the land, making the old man rich and hopefully resolving the feud. But the evil Red Baxter, owner of a saloon, tricks the boys out of the deed, and it's up to them - as well as Quale, who naturally finds his way out west anyway - to save the day.
After railroad agents forcibly evict the James family from their family farm, Jesse and Frank turn to banditry for revenge.
